Smac contains four contiguous amino acids, Ala-Val-Pro-Ile, that bind to its target proteins. Its Kd for binding is about 100 nM. The compound at right was found to bind to the same site on the same target protein with a Kd of 12 nM. Based on these data, and the compound’s structure, which of the four amino acids do you think is least important in binding to target proteins? Explain. B. The four amino acids listed above are at the N-terminus of Smac, and the 11 missing GFP amino acids are at the C-terminus. How do you suppose the binding of Smac to its targets is affected (if at all) when Smac binds to the remainder of GFP? Explain
