There are many ways by which an organism can become antibiotic resistant. some microbes have pumps on their cell surfaces( cell wall, cell membrane) which are effective in throughing out the antibiotics from inside the cell.

Many bacteria have plasmid(s) (extra- chromosomal material) which can contain the genes that codes for antibiotic resistance and they can transfer these genes to other bacteria which do not contain antibiotic resistant gene by horizontal gene transfer.

For example bacteria containing β-lactamase(enzyme) gene are resistant to penicillin(antibiotic) because penicillin contain β-lactam ring which is destroyed by the enzyme.
