Inside the cell, DNA polymerase cannot synthesize de novo (or “from scratch”): it can only add on to an existing 3’ end. So the cell uses the enzyme primase to first synthesize a short RNA primer, and DNA polymerase adds nucleotides onto the 3’ end of the RNA primer.
