The following shows the DNA recognition sequences for three restriction enzymes:

BglII: Sau3AI: BamHI:
5’...A^GATCT...3’ 5’...^GATC...3’ 5’...G^GATCC...3’
3’...TCTAG^A...5’ 3’...CTAG^...5’ 3’...CCTAG^G...5’

(A) You are working with a short linear double-stranded DNA fragment that has the sequence shown below. Make boxes that encompass the binding sites for each enzyme, and label them:

5 '3'
3 '5'

(B) How many fragments do you expect if you cut with BgII alone?
(C) Write out the sequences of each double-stranded fragment you will get after cutting with BamHI, Make sure to label the 5' and 3' ends of each strand of your fragments.
(D) How many fragments do you expect if you cut with BglII alone? How many fragments do you expect if you cut with Sau3AI alone? How many fragments do you expect if you cut with BamHI and BglII?
